Summary

The work was prepared in Chisinau in 2010 and has following structure: Introduction, six chapters, conclusions, recommendations, bibliography (308 sources), 3 annexes, 201 pages of basic text, 103 drawings, 19 tables. The results are published in 51 scientific papers, including 7 patents.

Field of research: relates to regenerative medicine, particularly in orthopedics-traumatology and surgery. The aim of the study is to highlight the properties of biological grafts (monocomponent and composite) with the cellular substrate as osteogenous remedies in disorder of strengthening bone.

Objectives: To study the possibilities of obtaining bone marrow cell grafts, cord blood stem cells and in vitro evaluation of the differentiation capacity of bone characteristic cells, proliferation ability and viability; assessment of the formalinized and demineralized bone grafts; development of a composite graft to optimize disturbed bone healing; the comparative study of the ability of cellular and composite grafts to heal the critical size long (radial), flat (parietal) and short (cuboid) bone defect in rabbits; to use these grafts in the treatment of pseudarthroses.

Novelty and scientific originality: were elucidated features of regeneration in experimental critical size defects in short, long and flat bones under the action of cell grafts and composite grafts. It was developed a device for administration of biological gelatin like drugs (MD 3556); a slide for cell culture research (MD 192 I2); it was proposed a pseudartroses treatment method, chronic amigdalitis and stress urinary incontinence by cell therapy (MD 3713, MD 3498, MD3313). It was developed a guide for collection, preservation and cultivation of the stem cells from ombilical cord blood.

Theoretical significance: consists in creating a concept for obtaining bone marrow and cord blood stem cells, and it’s use in areas of modern medicine, to create support for implementation of regenerative medicine methods in Moldova using advanced cellular technologies.

Applied value: еxperimental and clinical data obtained to assess cell and composite grafts are used to stimulate regeneration in non healed bones and other degenerative diseases. Determination of Biochemical Indicators of bone metabolism would enable early assessment of the effect of the graft in stimulating bone regeneration. The obtained results are used in students, residents and practical doctors trainings. Results implementation: the results were implemented in the research and teaching activity in SMPhU "Nicholas Testemitanu". The method was used in the treatment of pseudarthroses in 122 patients
