Summary

Purpose: To research the safety and pharmacological properties of a novel alkylisothiourea derivative – isopropylphosphite-S-isopropylisothiourea (profetur).

Objectives of the research: to determine specific (vasoconstrictor and hypertensive) effects and safety of isopropylphosphite-S-isopropylisothiourea; determination of changes of the main parameters of systemic hemodynamics and microcirculation in the application of profetur under normal conditions and on the background of experimental acute hypotension; investigation of the effect of the substance on the oxygen regime, acid-base and electrolyte balances of the body.

Scientific novelty and originality. For the first time it was investigated isopropylphosphite- S-isopropylisothiourea (profetur) - a new medicine derived from a series of antihypotensive isothiourea derivatives, were obtained data about its safety, its effects on systemic hemodynamics, microcirculation and oxygen consumption, the degree of oxygenation of the arterial and venous blood, acid-base and electrolyte balances in normal conditions and experimental drug-induced arterial hypotension.

Theoretical value. Isopropylphosphite-S-isopropylisothiourea proved to be harmless in laboratory animals. Single dose administration of profetur exhibits a pronounced and prolonged vasoconstrictor, hypertensive and antihypotensive actions, does not disturb essentially the systemic hemodynamics, microcirculation, oxygen regime, acid-base and electrolyte balances of the organism.

Practical value. Profetur can be recommended as antihypotensive drug for the prevention and treatment of acute hypotension, thereby increasing the number of vasoconstricting antihypotensive drugs of long action which are available for different routes of administration (intramuscular, intravenous) and at different stages of health care.

Implementation of scientific results. The results of research were implemented in the learning process at the Department of pharmacology and clinical pharmacology and the Department of pharmacology and clinical pharmacy, Departments of anesthesiology and reanimatology of the PI SUMP „Nicolae Testemitanu” from Republic of Moldova, in sections “Hypertensive drugs”, “Emergency medicines in allergic reactions, shock (traumatic, hypovolemic), poisoning with medicines etc.”.
